Long-Term Incubation of Cardiac Slices: Cardiac slices (125 µm thick) are prepared from mammalian left ventricle and mounted between a force transducer and length adjuster on the WolfPack system, with up to eight slices being mounted at the same time (Figure 1A). Slices are stretched to a sarcomere length of 2.2 µm, and isometric force transients for all eight slices are recorded simultaneously at multiple time points over a defined period of time (hours to days)(Figure 1B). This assay enables real-time monitoring of isometric transients, detection of long-term effects of compounds, and detection of real-time effects of gene therapies and other treatments.
